I have setup my 530 like described in the link. That should solve your issue (like dpawlyk and Wongataa mentioned).
If recalculation is disabled, the other settings don't matter at all.
The other settings only apply if you go off course and have the unit recalculate OR when you aren't using a course file (and have the unit calculate a route for you).
Except for "Lock on road". That just changes how the cursor (the blue triangle) is displayed (it doesn't change the route at all). If it's enabled, the cursor sticks to the nearest road. In my opinion, that not useful for cycling.

If you mean "course/route planner", it doesn't really matter (use what you prefer or find easiest). I use ridewithgps, which is capable and popular, and fairly easy to use.
The planners all produce the same data (which means they all will work the same for navigation).
The maps on the device are based on Openstreetmap (OSM). Most of the route planners let you use OSM maps. You are generally better-off using OSM map to plan your route.

Note that there are two separate forms of turn instructions: "turn guidance" *(the big white arrows) and "course points" (small indicators).
"Course points" are intended to be used on devices that don't use maps but the devices that use maps can display them.
You'll only get "course points" in FIT or TCX files. RWGPS adds them for every turn. Garmin Connect only adds your custom ones for devices that use maps.
Other than "course points", FIT, TCX, and GPX all work the same way (just make sure the file is a track).
