Does Garmin connect support offline mode? I need to sync new courses to my Garmin 25, and where I ride doesn't always have signal. My only alternative is to sync at home and leave the device turned on while I drive to my location.

If the Garmin 25 supports USB mass storage mode, the following (really cumbersome) procedure to transfer files from a phone may work:
Connect the device to an Android phone (that has USB OTG support) with a USB OTG cable (always via a powered USB hub!). The USB hub can be powered via a suitable adapter from the car's 12V. You may be able to copy the courses as files from the phone to the device.
But the way you do it now seems much easier.
