Loaded it twice. Selected Motorcycle, not direct. Routes are only direct. This software is useless if it won't follow roads, unless I got the airplane navigator by mistake :)
Since you have Motorcycle, not Direct selected, are you sure you have a map that's routable? BaseCamp world isn't, City Navigator is. OSM world maps are.
Hate to show my ignorance here, but I see a Profile labeled Direct, and when I choose Motorcycle I don't see Direct as an option. For that matter I don't see Direct as an option under any profiles. It might make sense for pedestrian, but it's not there either.
Nope. I'm the one who's not paying attention. The option to customize an activity using a "Direct" routing method has disappeared in the Mac version. I'm going to assume (bad idea) that the windows version has dropped it as well.

Almost none of the GPSFileDepot maps are capable of creating routes. Look at the download page for your map at GPSFileDepot. Unless it specifically says that the map is routable, it is NOT.
Some of the best things in life are free, but they take a little more effort.
If you want routing for roads only, spring for a copy of City Navigator. Can't be beat.
If you want a routable topo to view on BaseCamp you'll either have to buy a Garmin 24k topo or make it yourself.
For a handheld, if you are looking for a routable map of CO that includes trails, and is free, try this, and overlay a transparent topo on it. Preferably select a topo that doesn't include the information already drawn on the OSM map.
